Introduction. A tunable, pulsed mid-IR laser is focused on the sample’s surface. Photothermal induced resonance (PTIR) is a new technique which combines the chemical specificity of infrared (IR) spectroscopy with the lateral resolution of atomic force microscopy (AFM). Here, we demonstrate an epi-detected mid-infrared photothermal (epi-MIP) microscope at a spatial resolution of 0.65 μm. Optical Photothermal Infrared spectroscopy is a fast, non-contact optical technique that overcomes the IR diffraction limit. This causes photothermal effects, which are then measured using a visible probe laser. While at the same time, O-PTIR is not affected at all by fluorescence interference, a key limitation of Raman spectroscopy, thus allowing for submicron molecular characterization that is completely fluorescence free. O-PTIR microscopy overcomes several of the key limitations of traditional FTIR microscopy such as poor spatial resolution and complex sample preparation.
